Custom Reporter Cell Line & Reporter Assay Services

Reporter genes are crucial in biomedical research for tracking and quantifying cellular processes like gene expression and signaling pathways. Scientists use reporter assays to track the activity of regulatory sequences, transcription factors, and signaling pathways. This information is critical for elucidating disease mechanisms, identifying drug targets, and evaluating the efficacy and safety of potential therapeutics.

At Acres Biosciences, we specialize in custom reporter cell line generation and assay development services. Our advanced technologies and experienced team save you time and empower your research endeavors.

Luciferase reporter assay utilizes the enzymatic activity of luciferase enzymes, such as firefly luciferase, to measure gene expression or regulatory activity in cells. By coupling the luciferase gene with a target promoter region or regulatory sequence, researchers can monitor changes in gene expression in response to diverse stimuli or experimental conditions.

  • Capabilities: Customize transient or stable luciferase reporter cell lines tailored for dual luciferase reporter assay, celltiter glo assay, Promega HiBiT, and NanoBiT assay.
  • Applications: High-throughput screening, studies on transcriptional regulation, analyzing promoter activity, and exploration of signaling pathways.
  • Advantages: High sensitivity, wide dynamic range, and compatibility with high-throughput screening. Notably, dual luciferase reporter assay enables data normalization to internal control to eliminate experimental artifacts.

Fluorescent reporter technologies use fluorescent proteins like GFP or RFP to visualize cellular processes in real-time. By fusing these proteins to targets, researchers track protein dynamics and interactions.

  • Capabilities: Assays using fluorescent proteins like GFP and RFP, with options for customizing cell lines and assays.
  • Applications: Live-cell imaging and drug screening, which provides high-resolution insights into cellular dynamics.
  • Advantages: Non-invasive imaging, precise visualization, and compatibility with diverse platforms.

Beta-galactosidase reporter assays utilize the enzymatic activity of beta-galactosidase to detect gene expression or regulatory activity within cells. By linking the beta-galactosidase gene to a promoter region or regulatory sequence of interest, researchers can monitor changes in gene expression in response to different experimental conditions.

  • Capabilities: beta-galactosidase reporter assays.
  • Applications: Research on gene expression, promoter activity, and regulatory mechanisms in various cellular contexts.
  • Advantages: Simplicity, reliability, and ease of detection, making them versatile tools for basic research and drug discovery.Customize transient or stable luciferase reporter cell lines tailored for dual luciferase reporter assay, celltiter glo assay, Promega HiBiT, and NanoBiT assay.

Case Studies: Luciferase Reporter Cell Line & Luciferase Assay

HiBiT Assay

The HiBiT assay is a luminescent method used to quantify protein levels by detecting the HiBiT tag. It has been used for applications including protein quantitation, protein dynamics monitoring, and protein-protein interactions.

HiBiT stable cell line was generated by overexpressing target protein of interest with HiBiT. The cells were seeded into 384-well plate and treated with small molecule degraders for 16 hours. The luminescence was read by luminescence microplate reader.

Celltiter Glo Assay

Celltiter glo luminescent cell viability assay determines the number of viable cells in culture based on quantitation of the ATP present. It has been used for high-throughput screening (HTS), cell proliferation and cytotoxicity assays.

Cell viability was assessed using the CellTiter-Glo Luminescent Assay after 5 days of treatment with small compounds.
