T Cell Engagers (TCEs)
T-cell engagers (TCEs) are bispecific immunotherapeutics engineered to redirect endogenous T cells toward tumor cells by simultaneously binding a tumor-associated antigen and the CD3 receptor on T cells. This interaction forms an artificial immunological synapse that activates cytotoxic pathways and drives targeted tumor cell killing, independent of MHC restriction.

Dual binding assay
Evaluates TCE binding to both targets:
- Protein-based binding: ELISA
- Cell-based binding: ELISA, flow cytometry (FACS), SPR (Surface Plasmon Resonance), etc.
- Confirms simultaneous engagement of CD3⁺ T cells and target antigen–expressing cells

T cell activation assay
Measures T-cell activation and functional response through:
- Activation markers: CD69 and CD25 expression by flow cytometry
- Cytokine release: IFN-γ, TNF-α, IL-2, etc. (ELISA or multiplex platforms such as MSD/Luminex)
- Proliferation: CFSE or CellTrace Violet dye dilution
- Degranulation: CD107a mobilization and granzyme B release indicating functional activation

Cytotoxicity assay (TDCC)
Quantifies T-cell–mediated target cell killing through:
- Luciferase-based cytotoxicity assay (bioluminescence readout)
- Flow cytometry-based killing assay (e.g., 7-AAD, PI, Annexin V)
- LDH release assay for measuring target cell lysis

Mixed Lymphocyte Reaction (MLR)
Evaluates non–antigen-specific T-cell activation using allogeneic PBMCs through:
- Cytokine release analysis to assess off-target activation and cytokine-release risk
